演題1 「アグロバクテリウム植物相互作用のシステム生物学」
Systems biology of Agrobacterium plant interaction

 土壌細菌のAgrobacterium tumefaciensは植物を遺伝的に形質転換させるユニークな機構を持つ。細菌のTi(tumor inducing:腫瘍誘導)プラスミドの一部分は受容する植物細胞に転移し、染色体ゲノムに組み込まれる。細菌遺伝子の発現により、感染細胞は腫瘍のような増殖を示す。
 この腫瘍形成機構はモデル植物Arabidopsis thalianaを用いて主に研究が進められ、細菌と植物のどのような遺伝子が細菌DNAの転移と組み込みに必要なのか明らかにされた。T-DNA(転移DNA)はTiプラスミドの一部であり、細菌側の特別な転移装置により、一本鎖DNAの状態で植物側の受容細胞に移動する。'ビルレンスタンパク質'と呼ばれる特別なタンパク質群はこのT-DNA末端に付着し、あるものは全体に渡って結合する。植物特異的なタンパク質と協調して、ビルレンスタンパク質はT-DNA/タンパク質複合体が核膜孔を通って核内部へ移動するのを可能にしている。しかし、この後の組み込み機構はほんのわずかしか分かっていない。
 T-DNA上にコードされている遺伝子群の解析から、腫瘍形成遺伝子を除いた細菌ベクターが開発された。これによって、稔性のある植物に再生しうる形質転換細胞の開発ができるようになった。植物種が異なれば、形質転換能は異なり、いくつかの植物では形質転換を行うことは今でも困難である。形質転換植物の利用は植物遺伝子の機能解析に供されるだけでなく、農業上利用価値の高い穀物を創生する基盤技術でもある。

 1980年、妻(バーバラ・ホーン)と一緒に進めて来たバクテリオファージの研究に区切りをつけて、我々は真核生物と植物ウイルスにテーマを変えることにした。我々はカリフラワー・モザイク・ウイルスを選んだ。当時、このウイルスはDNAを持ち、植物細胞質に形成される封入体(inclusion body) に蓄積することが分かっていた。我々はこのゲノムがレトロウイルスのように逆転写酵素によって複製するが、それとは違い、核内にミニ染色体を形成し、核ゲノムに組み込まれないことを確かめた。
 しばらくして、我々はウイルスを遺伝子ベクターに改変し、例えば、インターフェロン遺伝子を植物で発現させることに成功した。
 その後、ウイルスの酵素や構造タンパク質をコードする7つのオープン・リーディング・フレームの機能を解析した。これらの一つであるtransactivator/viroplasmin (TAV)は多頭の竜のように、封入体マトリックスタンパク質、ウイルスが集合するときの触媒因子、核とのシャトルタンパク質、ポリシストロニックな翻訳のトランスアクチベーター、免疫エフェクター (immune-effector)、そして特に、サイレンシング・サプレッサーとしての機能を持つことが判明した。
 ウイルスRNAは複製中間体として働き、驚くべきことではないが、それはポリシストロニックであり、翻訳のときにTAVを必要とする。このことは一旦翻訳が終わっても引き続き翻訳の開始 (reinitiation) を可能にする興味深い機構を示している。さらに、このRNAは非常に複雑なリーダーから始まり、リボゾームはスキャニングするとき、この部分を迂回 (shunt) する。このリーダーからは短いRNAも発現し、このRNAは二本鎖に変換され、莫大な量のsiRNAのもとになる。これらは'おとり (decoy)'としての役割があり、RNAに基づくサイレンシング・サプレッサーで、Argonauteタンパク質の作用をブロックする働きを持つ。

 バーバラ・ホーン博士とトーマス・ホーン博士はお二人ともに1960年代後半にバクテリオファージから研究をスタートされました。その後、アグロバクテリウムを用いた植物分子生物学と植物ウイルス学にそれぞれシフトし、以来これら学問分野の基礎から応用開発まで幅広い領域を、文字通りトップランナーとして牽引されてきました。
 いうまでもなく、ホーン夫妻は世界的に著名な科学者であり、今回、国際ワークショップ (Plant Genome Stability and Change 2016, Hayama, July 7-10, 2016)と国際シンポジウム (Frontier in Plant Genome Editing, Tokyo, July 10-11, 2016) での基調講演のために来日の予定です。本学でもご講演をしていただくことになりましたので、アグロバクテリウムや植物ウイルスを含めた植物科学 (Plant Science) にご関心・ご興味をお持ちの皆様のご来聴を心より歓迎します。


演題1
  The bacterium Agrobacterium tumefaciens uses a unique mechanism to genetically transform plants. A portion of the bacterium's Ti (tumor inducing) plasmid in transferred to recipient plant cells, where it integrates into chromosomal DNA. Expression of bacterial genes results in tumorous growth of the affected plant cells.
  Study of this mechanism of tumorigenesis, mainly carried out in the model organism Arabidopsis thaliana, revealed the requirements for bacterial and plant genes that enable the transfer and integration of the bacterial DNA. The T-DNA (for transferred DNA) is part of the Ti plasmid and moves to the plant recipient in single-stranded form, using a special transfer apparatus of bacterial origin. Dedicated bacterial proteins called virulence proteins attach to this T-DNA and bind along the whole length, respectively. In conjunction with plant-specific proteins they also permit the passage of the T-DNA/protein complex through the nuclear pore into the nuclear compartment. The subsequent integration event is only partially understood.
  Analysis of T-DNA located genes led to the development of bacterial vectors that are devoid of tumor-forming genes and that thereby permit the establishment of transgenic cells that are capable of regeneration to mature, fertile plants. Different plant species exhibit different levels of permissiveness for transgenesis with some plants being rather recalcitrant. The use of transgenic plants offers to study functions of plant genes. In addition, it also is the basis of generation of crop plants of agricultural value.

演題2
  1980, after having worked together with my wife on bacteriophage, we decided to switch to eukaryotes and a plant virus. We choose Cauliflower mosaic virus, which was known to contain DNA and accumulate in "inclusion bodies" within the plant cytoplasm. In a sudden light bulb moment, we recognized that its genome replicates by reverse transcription, like retroviruses, but unlike those, it establishes itself as nuclear mini-chromosome and does not integrate.
  Later-on, we transformed the virus into a gene vector, carrying, for instance, an interferon gene to be expressed in plants.
  One after the other, we characterized the functions of its seven open reading frames, coding for enzymes and structural proteins. One of these proteins, "transactivator/viroplasmin" (TAV), turned out to be a multi-headed dragon, functioning as inclusion body matrix protein, catalysator of virus assembly, nuclear shuttle protein, transactivator of polycistronic translation, immune-effector and, last not least, silencing suppressor.
  The virus RNA serves as a replicative intermediate, and, not surprisingly it is polycistronic and requires TAV for translation, revealing an interesting mechanism that allows reinitiation of translation. Furthermore, this RNA starts with a very complex leader, which is shuttled by the scanning ribosome. This leader is also expressed as a short RNA, which becomes double-stranded and give rise to massive amounts of siRNAs. these act as decoys, RNA-based silencing suppressors, blocking the action of Argonaut.
